Lens-free microscopic technology utilizes Gabor-based in-line holographic optical path to capture the interference images generated by illuminating lights through the micro-samples, with the help of an area array detector but without any imaging lenses. It is a digital microscopic imaging technology which uses a digital-picture-processing method to reconstruct image for the sake of acquiring information from these micro-samples. The shrinkage of equivalent pixel elements achieved by pixel super-resolution gives more high-frequency information and, further, it promotes the resolution of reconstruction directly. Meanwhile, multiple phase recovery arithmetic tools make the same contributions to resolution indirectly by eliminating the twin-images, especially to the dense samples. Moreover, such technology breaks the limitation in spatial bandwidth product, comparing to the classical optics microscopes, which leads to the high-resolution imaging in a larger field of view (FOV). Therefore, it can provide powerful supports for rapidly diagnosing samples with a big field of view in clinical application and examination in the case of resource shortage. Additionally, the booming algorithm with optimizing hardwares renders impetus in data-sampling and computing effects, expanding the applications on specimens moving in high speed and on a nanometer scale. The developing tendency of lens-free technology and its matched hard devices are to become compacted both arithmetically and volumetrically. Heterologous Expression and Activity of Rhamnolipid Gene Regulated by Three Different Constitutive Promoters in Pseudomonas protegens

In this research, the recombinant expression vector pBBR1-genta-RhlAB containing the rhamnosyltransferase gene (RhlAB)was modified via Red/ET homologous recombineering. Then, three recombinant expression vectors pBBR1-genta-Papra-RhlAB, pBBR1-kan-Ptac-RhlAB and pBBR1-kan-PrhaB-RhlAB with three different constitutive prIn this research, the recombinant expression vector pBBR1-genta-RhlAB containing the rhamnosyltransferase gene (RhlAB)was modified via Red/ET homologous recombineering. Then, three recombinant expression vectors pBBR1-genta-Papra-RhlAB, pBBR1-kan-Ptac-RhlAB and pBBR1-kan-PrhaB-RhlAB with three different constitutive promoters (Papra、Ptac and PrhaB)were constructed by subcloning technology, and then we electro-transformated the resultant expression vectors into P.protegens Pf-5.When the flask in LB medium was fermented to 42 h, the rhamnolipid concentration of Pf-5/pBBR1-genta-RhlAB supernatant was 17.56 mg/L, with that of Pf-5/pBBR1-genta-Papra-RhlAB, Pf-5/pBBR1-kan-Ptac-RhlAB and Pf-5/pBBR1-kan-PrhaB-RhlAB supernatant being 11.135 mg/L, 441.135 mg/L and 557.764 mg/L, resulting in 0.63, 25.12 and 31.76 fold variation after promoter optimization. High performance liquid chromatography-mass spectrometry (LC-MS/MS)analysis showed that there were total 4 rhamnolipid derivatives in the Pf-5 engineering bacteria. Furthermore, the RhlAB gene expression was later examined by qRT-PCR with the result showed that the expression level of RhlAB gene after Ptac and PrhaB promoter was 2.16 and 2.77 fold higher than that of the original one. Generally, this study preliminarily realized the expression of RhlAB gene in Pf-5, and found that the constitutive promoter Ptac and PrhaB was more efficient than the original promoter which could provide a significant reference with significance for heterologous biosynthesis of rhamnolipid..

Real-Time Monitoring of Viscoelastic Changes of Neonatal Rat Primary Cardiomyocytes Subjected to the Treatments of Different Concentrations of Inotropic Drugs Using Quartz Crystal Microbalance

The adhesion of neonatal rat primary cardiomyocytes onto gold electrode surface, and the subsequent drug effects of different concentrations of cardiomyocyte positive inotropic drug isoprenaline and cardiomyocyte negative inotropic drug verapamil, were monitored through the frequency (F)and motional resistance (R)measurements of the QCM in a real-time way. Cell viscoelastic index CVI (CVI=ΔR/ΔF)was used to characterize the viscoelastic changes of the cells, and the drug-induced morphological changes of the cells were observed by optical microscope. The results showed that with the increase of isoprenaline concentration, the magnitudes of QCM frequency drop, R increase and CVI increase all increased; whereas with the increase of verapamil concentration, the magnitudes of QCM frequency increase, drops in both R and CVI all increased. Cells’ contraction under the treatment of isoprenaline was observed under optical microscope, which is consistent with the result of increased CVI indicating that the cells became stiffer; while cells’ relaxation under the treatment of verapamil was observed under optical microscope, which is consistent with the result of decreased CVI indicating that the cells became softer. Our results suggest that the QCM has a great application potential as a tool in cardiovascular drug screening and evaluations..

Establishment and Characteristics Analysis of in vitro Culture An Ear Fibroblast Cell Line Derived from Hartley Guinea Pig

This experiment was performed to rapidly obtain fibroblasts from guinea pig’s ears, and provide initiating cells for somatic cell reprogramming. The ear tissues were minced and digested with 0.05% Trypsin-EDTA and Collagenase IV. The cells were cultured in vitro and morphologically observed. The cells were detected for mycoplasma and vimentin protein immunofluorescence was performed for characterization. The cell growth curve was drawn to calculate the cell double time. And the infection efficiency was detected by flow cytometry when the cells were infected with retrovirus containing green fluorescent protein. The influence of the virus on the karyotype was detected by karyotype identification. The isolated guinea pig ear fibroblasts were adherent growth cells. Most of them were adherent and fully extended 2~3 hours after digestion. The cells were fusiform or polygonal, with full and stereoscopic cytoplasm. And the nucleus was clear and in good shape. The mycoplasma detection was negative. The cells expressed fibroblast-specific protein vimentin. The growth curve was S-shaped, which was consistent with the normal growth and proliferation rules of cultured cells in vitro. The cell double time was 24.85 h. The efficiency of retrovirus infection was more than 70%, and the cells with chromosome 2n=64 accounted for 90% after viral infection, with genetic stability and normal karyotype. The method is simple and efficient, producing rapidly proliferated cells in good condition. It is an appropriate method for isolating fibroblasts from guinea pig’s ears. The cell line has a high virus infection rate, which can be used for somatic cell reprogramming studies..

The ISSR Analysis of 24 Dendrobium officinale Germplasm Resources

The genetic diversity and genetic relationship of 24 Dendrobium candidum germplasms with different yields were studied by ISSR molecular markers in this study. Seven ISSR primers were screened to amplify genomic DNA of Dendrobium officinale by PCR. The genetic similarity (GS)of twenty four varieties by NTSYS-pc software ranged from 0.478 3 to 0.913 0. Cluster analysis showed that 24 cultivated populations of Dendrobium officinale could be divided into three groups. The results showed that the genetic diversity of Dendrobium officinale germplasm resources was abundant. It provides a reliable theoretical basis for comprehensively understanding the genetic background, the selection, introduction and promotion of Dendrobium officinale in this research..

Construction of Zebrafish gpr98 Gene Knockout Experimental Model

Constructing the gpr98 gene mutant strain of zebrafish can provide an excellent animal model and research basis for explaining the function of the gpr98 gene. In this paper, two target sites with a distance of 42 bp in exgpr98 is associated with a variety of diseases, and it is highly conserved in human and zebrafish. Constructing the gpr98 gene mutant strain of zebrafish can provide an excellent animal model and research basis for explaining the function of the gpr98 gene. In this paper, two target sites with a distance of 42 bp in exon 2 of zebrafish gpr98 gene were selected for sgRNA recognition. SgRNA was synthesized in vitro and co-injected with Cas9 mRNA into the embryo of zebrafish at 1-cell stage. A few partial embryos were randomly selected for DNA genotyping when they are in 72-hour stage. The results showed that in addition to the wild-type DNA bands, some embryos have a band smaller than the wild-type DNA bands. The F0 positive individuals were mated with wild-type zebrafish, and the offspring of the F0 generation positive individuals were tested for genotyping, and a stable genetic mutation strain with a deletion of 48 bp was obtained. Mutation Rates at 18 X-chromosome STRs in the North China Han Population

Genomic DNA samples extracted from blood stains from 484 lineage members from North Chinese Han population were genotyped with TYPERX19 kit. In a total of 5 652 allele transmissions, 8 mutations were detected from 6 different markers including DXS6810, DXS8378, DXS6797, DXS7423, DXS7424, DXS8377 through the 484 lineageGenomic DNA samples extracted from blood stains from 484 lineage members from North Chinese Han population were genotyped with TYPERX19 kit. In a total of 5 652 allele transmissions, 8 mutations were detected from 6 different markers including DXS6810, DXS8378, DXS6797, DXS7423, DXS7424, DXS8377 through the 484 lineage members, the mutation rate of DXS6810 and DXS8377 was 0.006 4, the mutation rate of DXS8378, DXS6797, DXS7423 and DXS7424 was 0.003 2. No mutation was found in the other 12 markers. The average mutation rate across all 18 X-STR markers was 0.001 4. All mutations were with single step. There were no significant differences between mutation rates of different X-STR loci (P>0.05). The losses and gains were balanced at all loci. Mutation rates were correlated with the lengths of the alleles. Alleles with higher number of repeats were more likely to mutate. Screening and Agronomic Analysis of Mutants from Indica Maintainer Rice T98B Treated with γ-Ray Irradiation

Radiation mutagenesis is available for crops germplasm creation. In this study, gamma-ray irradiation was used to induce the rice cultivar T98B which is a main maintainer line of hybrid rice, under the treatment of mature seeds at a dose of 300 Gy. It focused on screening and grouping of agronomic mutants, and the freqRadiation mutagenesis is available for crops germplasm creation. In this study, gamma-ray irradiation was used to induce the rice cultivar T98B which is a main maintainer line of hybrid rice, under the treatment of mature seeds at a dose of 300 Gy. It focused on screening and grouping of agronomic mutants, and the frequency for each type was also investigated. The results showed that, by contrast of T98B, the germination rate for M1 plants and M2 plants were both significantly suppressed, with a decrease of 22.35% and 14.67% respectively. In M2 population, 207 candidate mutants were screened. Of which, 168 were validated steadily inheritable via a phenotyping test from M3 to M5, accounting for a total mutation frequency of 0.673%. Furthermore, they were divided into 3 groups regarding leaf group, stem/tiller group and grain/flower group, according to the part that it occurred to mutate on. Each group above included 34, 37 and 97 mutants respectively, representing a ratio of 20.24%, 20.02% and 57.74%. The common mutants were the ones varied on fertility, leaf color and plant height, and it presented a mutation frequency of 0.212%,0.092% and 0.088% respectively. Moreover, a brief description on the phenotypes for each group was provided. The results indicated that, agronomic mutants in rice could be effectively obtained via gamma-ray irradiation, and several mutants gained here would be severed as useful germ resources for advancing the work on rice genetic improvement..

A Study on the Effects of Cultivation Factors on Cotton Yield in Short Season

The seeding period has obvious negative effect on seed cotton yield, and nitrogen fertilizer and density have obvious positive effect on seed cotton yield. The order in which the two factors interact on the yield of seed cotton is as follows:the interaction effectsof nitrogen and planting density>density and seeding period>Nitrogen application and sowing date.
